Abstract

Rainbow tree leaves (Eucalyptus deglupta Blume) are plants known to contain various secondary metabolite compounds such as flavonoids, alkaloids, saponins, steroids, and carotenoids that have potential as natural antibacterials. Hand sanitizer gel made from natural ingredients is an interesting alternative to reduce the spread of pathogenic microorganisms on hands. This study aims to formulate and test the antibacterial activity of hand sanitizer gel based on rainbow tree leaf extract against Staphylococcus aureus bacteria. The extraction was carried out using the maceration method with 96% ethanol as the solvent, then formulated into gel preparations with three variations of extract concentrations (5%, 7.5%, and 10%). using Carbopol 940 as a gelling agent. Evaluation was carried out on the physical properties of the gel, including organoleptic properties, pH, homogeneity, spreadability, adhesion, and viscosity, as well as antibacterial activity testing using the agar diffusion method. The results showed that all formulas had good physical characteristics and an appropriate pH for the skin. Antibacterial activity increased with the concentration of the extract, where the 10% formula produced the largest inhibition zone (9.0 mm), exceeding the positive control (6.7 mm). Therefore, the rainbow leaf extract has the potential as a natural active ingredient in hand sanitizer gel formulations to inhibit the growth of Staphylococcus aureus.
